Model reduction for ‐induced vesicle fusion dynamics

چکیده

Abstract In this work, we adapt an established model for the ‐induced fusion dynamics of synaptic vesicles and employ a lumping method to reduce its complexity. reduced system, sequential ‐binding steps are merged single releasable state, while keeping important dependence reaction rates on local concentration. We examine feasibility reduction representative stimulus train over physiologically relevant site‐channel distances. Our findings show that approximation error is generally small exhibits interesting nonlinear non‐monotonic behavior where it vanishes very low distances insignificant at intermediary Furthermore, give expressions model's suggest our approach may be used directly compute effective assessing validity model, thereby circumventing expensive simulations.

-dependent Vesicle–Vesicle Fusion Events

A microneedle puncture of the fibroblast or sea urchin egg surface rapidly evokes a localized exocytotic reaction that may be required for the rapid resealing that follows this breach in plasma membrane integrity (Steinhardt, R.A,. G. Bi, and J.M. Alderton. 1994. Science (Wash. DC). 263:390–393). How this exocytotic reaction facilitates the resealing process is unknown. We found that starfish o...

Molecular dynamics simulations of lipid vesicle fusion in atomic detail.

The fusion of a membrane-bounded vesicle with a target membrane is a key step in intracellular trafficking, exocytosis, and drug delivery. Molecular dynamics simulations have been used to study the fusion of small unilamellar vesicles composed of a dipalmitoyl-phosphatidylcholine (DPPC)/palmitic acid 1:2 mixture in atomic detail. The simulations were performed at 350-370 K and mimicked the temp...

Complexin synchronizes primed vesicle exocytosis and regulates fusion pore dynamics

ComplexinII (CpxII) and SynaptotagminI (SytI) have been implicated in regulating the function of SNARE proteins in exocytosis, but their precise mode of action and potential interplay have remained unknown. In this paper, we show that CpxII increases Ca(2+)-triggered vesicle exocytosis and accelerates its secretory rates, providing two independent, but synergistic, functions to enhance synchron...

Computational model for autophagic vesicle dynamics in single cells

Macroautophagy (autophagy) is a cellular recycling program essential for homeostasis and survival during cytotoxic stress. This process, which has an emerging role in disease etiology and treatment, is executed in four stages through the coordinated action of more than 30 proteins. An effective strategy for studying complicated cellular processes, such as autophagy, involves the construction an...
